No! Travelling to Puerto Rico is within the U.S. so as long as you are a U.S. citizen, no passport is needed.
No, you don't need to know Spanish, although we always recommend learning the basics ;) Most people speak English in Puerto Rico so you should be just fine not knowing Spanish. Many of the street signs may be in Spanish. They do use the metric system so you will see signs for kilometers, gas by the liter, etc. We recommend changing your navigation system and car settings before driving to easily be able to navigate. If you would like to be able to easily translate road signs, menus, etc. you can use Google Translate.
While the venue is pretty close to San Juan, the traffic pattern is constantly changing within the city, especially in Old San Juan or Viejo San Juan so we recommend visiting that area on another day and avoid going into to San Juan that day. That being said, if you stay on the East side of San Juan or outside of the city (check out our travel section later for recommendations), you should be fine to plan for 30 minutes to an hour drive.
